Arrowhead Pharmaceuticals Advances in Obesity Treatment with Gene-Silencing Candidates

Arrowhead Pharmaceuticals has reported early-phase data indicating that its gene-silencing drug candidates may aid in weight loss for individuals with obesity and Type 2 diabetes. In a Phase 1/2 study, participants receiving ARO-INHBE, a candidate targeting the INHBE gene, in combination with Eli Lilly’s GLP-1/GIP receptor agonist tirzepatide, experienced a weight loss of 9.4% over 16 weeks. In contrast, those on tirzepatide alone lost 4.8%. Additionally, the combination therapy led to a 23% reduction in visceral fat, a 15% decrease in total fat, and a 77% reduction in liver fat, which is significant given the health risks associated with excess liver fat.

Arrowhead’s ARO-INHBE is designed to reduce hepatic expression of the INHBE gene, while another candidate, ARO-ALK7, targets the ACVR1C gene to decrease adipocyte expression. However, the data on ARO-INHBE's efficacy as a standalone treatment remains limited, as Arrowhead has not disclosed monotherapy weight loss results.
